...Which one is the best?

Which ones do you use (if any)? Can they play multiple parts at the same time?

And are there any that can "play" other instruments?

the best? PowerTab...makes real professional qulaity sheet music, shows actual notes as well as tab, and has sooo many functions. Guitar pro is easier for making MIDIS and such but Ive always liked PowerTab better.

guitar pro can play multiple tracks, and has alot of instruments to choose from, thats why its easier to make MIDIs on, I write my symphonic pieces on guitar pro, but as I mentioned earlier for guitar stuff, I go PowerTab.
