So I'm building a polka dot vee copy just like Randy's. I'm making the body out of mahogany like his and using a vintage Danelectro neck like his had.

I got the Dano neck but I need to buy a 6 screw tremolo.

Does anyone know how I determine the string spacing based on the measurements of the neck? I think it goes according to the heel width right?
 
You need to layout a full scale mockup of the nut, strings and bridge.

You know the nut width. A Strat bridge has 2 7/32" to 2 3/8" spacing.

Put the high E saddle the same distance that it is from the 12th fret to the nut. Leave enough room for the 1/2" +/- setback for the low E compensation. Hopefully this will give you around 1/8" clearance from the sides of both E strings to the edge of the neck. If it isn't Gibson bridges are around 2 1/16" and I have an old Kahler that is under 2".

After all that you will have to figure out a good spot to place that body/neck joint.

Always best to draw things out and double check, but from my experience:

Measure the heel width.

Your bridge string spacings usually vary between 2 1/16, 2 1/8 & 2 7/32.
If the heel is 2 1/4, any of those will work, you'd just have slightly more room on either side of the e strings with the narrower bridge, but it won't be excessive.
If the heel is smaller than 2 1/4, I'd probably just avoid the 2 7/32. I think the strings are too close to the edges of the neck with that setup.
