how to measure reverb in guitar for a show ?

This is a "live prod" question hehe

I have my GT-10 and I can set the time of my Hall2 reverb... 2.4s (level 66) with a hiss cut of 4khz seems right for the rehearsal room, but we have a show where like about 400 person can enter, how should I set the reverb ?

If it sounds good it is good. You don't need to adjust it for the show. The FOH will mic it and make it louder.

The reverb in the room changes but the reverb yo're using is a separate thing. There are certainly some venues where you don't need any extra reverb but in practice I don't know any touring guys that adjust all of their presets based on the venue.
