4x12 vs. two 2x12s?

Are two 2x12s going to sound basically the same as a single 4x12 if speakers, wood thickness, construction, and internal volume are all the same? I've never been able to A/B two comparable setups so I don't have much sense of what the similarities or differences are...

You mean there's less low end overall? How is the directionality affected?

The reason I'm asking is that I'm currently using a pair of 2x12s that I build myself (baltic birch, v30s, pretty standard) but occasionally I'll think about getting an empty 4x12 and transplanting the speakers...

There is a difference between a 4x12 and a "split 4x12" 2x12 cab. Even though the construction is the same and the 2x12 has exactly half the internal volume as the 4x12 it doesn't sound quite the same because of the interaction of the 4 speakers in the 4x12. You will note that 2x12's that are labeld as "sounds like a 4x12" are larger in size than a "split 4x12" 2x12 cab.
